Applications are invited for the post of Research Associate in the Photonics Group, Department of Physics, based at the South Kensington Campus of Imperial College London.

This is a full-time post funded by an Imperial Confidence in Concept & AstraZeneca Innovation Fund award led by Dr Chris Dunsby at Imperial College London and Dr Delyan Ivanov and Dr Radek Polanski at AZ. The aim of this project is to develop and apply a unique single-objective light sheet microscopy system – an oblique plane microscope (OPM) – to automated medium-throughput imaging of multicellular spheroids for applications in compound safety screening. The drug discovery pipeline needs accurate information on the toxicity profile of potential drug candidates. Currently, the vast majority of this work is performed using conventional 2D/monolayer cell culture but it is becoming increasingly clear that this does not adequately reflect the complexity of real tissue. This leads to unwanted toxicity and drugs failing at late stages in the pipeline due to safety concerns that were not detected in earlier tests. This project aims to test whether OPM can provide insight into the mechanism of toxicity and into the cellular health of cells within the spheroid core.

You will be responsible for the design, construction, testing and application of optical systems for light sheet fluorescence microscopy. You will also develop and apply software to control automated imaging, including 3D imaging in multiwell plates, to achieve automated 3D image analysis and quantification of large time-lapse 3D datasets. You will also work closely with collaborators at AZ to develop sample preparation strategies and to image samples.

You must have a degree (or equivalent) in physics, engineering or similar and a PhD (or equivalent) in optical science including fluorescence imaging and software development for signal/image processing. You must also have significant experience of the development and application of fluorescence imaging systems, and will be proficient in the alignment of complex optical imaging systems. You will also have experience of software development for computer control of experiments and for 3D image processing and analysis including the use of LabVIEW and MATLAB. Knowledge of 3D fluorescence imaging techniques such as confocal and light sheet microscopy is also essential.
